Two things to check;

    - Make certain the group ownership of your mailman directory trees
      and their contents are owned by the same user/group that your Mailman
      daemon runs as.

    - You may have odd perms set on that queue file you mention.
      I don't believe the 'check_perms' will modify perms on queue
      files.

Here is what my Mailman tree looks like;

    drwxrwsr-x   19 root     mailman      4096 Jan 31 22:15 .
    drwxr-xr-x   25 root     root         4096 Feb 29 04:58 ..
    drwxrwsr-x    4 root     mailman      4096 Jan 29 13:50 archives
    drwxrwsr-x    2 root     mailman      4096 Feb 17 17:00 bin
    drwxrwsr-x    2 root     mailman      4096 Jan 29 13:59 cgi-bin
    drwxrwsr-x    2 root     mailman      4096 Feb 16 11:50 cron
    drwxrwsr-x    2 root     mailman      4096 Mar  7 22:54 data
    drwxrwsr-x    8 root     mailman      4096 Feb  3 14:11 lists
    drwxrwsr-x    2 root     mailman      4096 Mar  9 07:15 locks
    drwxrwsr-x    2 root     mailman      4096 Jan 29 13:50 logs
    drwxrwsr-x    2 root     mailman      4096 Jan 29 13:59 mail
    drwxrwsr-x   11 root     mailman      4096 Feb 16 11:48 Mailman
    drwxrwsr-x   28 root     mailman      4096 Jan 29 13:51 messages
    drwxrwsr-x    6 root     mailman      4096 Jan 29 13:59 pythonlib
    drwxrwsr-x   11 root     mailman      4096 Jan 29 13:50 qfiles
    drwxrwsr-x    2 root     mailman      4096 Feb 16 11:49 scripts
    drwxrwsr-x    2 root     mailman      4096 Jan 29 13:50 spam
    drwxrwsr-x   30 root     mailman      4096 Jan 29 13:51 templates
    drwxrwsr-x    4 root     mailman      4096 Feb 16 11:48 tests

Let us know how it works out.
I remember upgrading from MM 2.0.13 to 2.1.4 and it surely wasn't fun
by any means - so you're no the only one to have issues ;-)
